A. THE EFFECT OF ULTRAVIOLT RADIATION ON BACTERIA Objectives 1. Students should be able to explain the deleterious effects of ultraviolet radiation on the growth and development of bacteria. 2. Students should be able to describe the effect of ozone on ultraviolet radiation. MATERIALS 1- Broth culture of Escherichia coll. 2- Three nutrient agar plates. 3-Ultraviolet (germicidal) lamp 4-Carboard or paper mask with design to cover the bottom half of the Petri dish. МЕТНODS 1. Inoculate all three plates with E. coli by spread plating technique using a bent glass rod or a sterile swab stick to produce a lawn of bacterial cells on agar. Do not use a streak technique. 2-Cover the bottom half (take the cover ofl) of two Petri dishes with cardboard or paper, and keep the third plate covered with the lip. 3-Radiate all three plates directly under the lamp. Do not look at the lamp. Expose one plate (with paper design) for 5 minutes and second plate for 10 minutes and third plate with lid in place also for 10 minutes.
