A cell is placed in a medium with volume equal to that of the cell. Then 100 marked molecules are placed inside, and after 1 h, 40 are found inside and 60 are found outside. In a control, protein in the membrane thought to be involved in transporting the molecule has been removed and 50 out of 100 of the molecules are found inside after the same amount of time.
