2. 2 (a). list Examine the methodologies given below and five consumables/chemicals/materials that require sterilization. Choose the best sterilization method for each listed consumable/chemical/material and explain your reasons for choosing the sterilization methods. The methodologies for culturing Escherichia coli in a chemostat (modified from Tonoyan, L., Fleming, G. T., Friel, R., & O'Flaherty, V. (2019). Continuous culture of Escherichia coli, under selective pressure by a novel antimicrobial complex, does not result in development of resistance. Scientific reports, 9(1), 1-11) The continuous culture of E. coli was conducted in a glass culture vessel, which is the chemostat, containing lysogeny broth (LB) medium as shown in Figure Q.2.(a). The chemostat vessel was submerged in a temperature-controlled water bath operated at 37C. The cultures were mixed by bubbling with air. The fresh media was introduced at a constant flow rate by using a peristaltic pump and the culture waste was removed at the same rate from the culture vessel. Samples were taken from the chemostat using a syringe attached to the sampling port.
