2. Select one isolated colony for transfer from your UMO plate, ideally from the 3rd streak Follow aseptic technique procedures! Gently pick up one colony off your plate, being careful not to mix it with other colonies around it. Use aseptic technique for each transfer. Move the colony to an agar slant tube. See appendix 4 if you need some help with this transfer. After incubation, observe your new tube to verify growth. Periodically you will be making a new slant tube for your UMO in order to maintain its growth. Follow the schedule of your instructor for when to remake the slant tube
