An HPLC experiment was performed to determine the amount of butyl paraben preservative in a commercial sample of hand lotion by direct calculation external standard method. The allowable limits of the preservative in these lotions has been set by Health Canada at 0.1% - 0.5% wt/wt. The following data was obtained:

Sample preparation: 1.038 grams of paste was quantitatively transferred to a 250ml volumetric flask, 50mls of M.P was added to the flask and the flask was sonicated then shaken to extract the preservative. The sample was then diluted to volume with remaining mobile phase. This solution was then passed through a 0.45 um filter and the first 2-3 mls was discarded. A subsequent portion was then collected in a 50ml beaker for injection into the HPLC system.

Standard preparation: 170.5 mg of butyl paraben reference standard was quantitatively transferred to a 200ml volumetric flask. 50 mls of MP was added to dissolve the standard, and then it was diluted to volume. 5.0 mls of this stock standard was diluted to 200mls V.F with M.P. This solution was then passed through a 0.45 um filter and the first 2-3 mls was discarded. A subsequent portion was then collected in a 50ml beaker for injection into the HPLC system.
