Your grade for this experiment will consider procedure, not just the identity of the microbe. Think Dichotomously and only do the necessary tests! Slide Coagulase directions: loop. 1. Emulsify a visible amount of the B organism in a drop of dH20 with a 2. Add 2 loops of coagulase (rabbit) plasma. 3. Stir with your loop for up to 1 minute and look for clumping. PYR Kit directions: 1. Place disc on clean slide. Use 1 loop of dH20 to moisten and hold on the slide. 2. Smear a visible amount of the microbe on the disk with a sterile loop. 3. Wait 5 minutes. 4. Add 1 drop PEP reagent. Read after 1 minute, but not longer than 2 minutes for a bright, hot pink color of the disc. When doing a catalase test from a blood agar plate, do NOT scoop part of the media along with the microbe since RBCs have catalase. You may get a false + reaction. For disc diffusion tests, use confluent growth of the microbe on the appropriate plate. See the experiment for instructions. After incubating the plates, measure any zones of inhibition and compare to the standards given in this experiment to determine whether the microbe is resistant or susceptible. 199
