1. Stock reagents: Solution A:1% w/v copper sulfate (CuSO4SH2O) Solution B: 2%w/v sodium potassium tartrate Solution C: 0.2MNaOH Solution D: 4% Sodium carbonate ation of rrotein Concentration Lowry reagent II (Folin-Ciocalteu 2x reagent) 2. To 49mL of Solution C add 49mL of Solution D. Then add 1mL of solution A and 1mL of Solution B. This is the copper-alkali reagent (Lowry Reagent I), which must be prepared freshly for each lab session. 3. Dilute the Folin-Ciocalteu reagent 2-fold (i.e. dilute with an equal volume of water). This is Lowry reagent II. 1. Make up a set of standards using BSA or some other protein source in the range of 5500g/mL. In the Lowry procedure, you will use 0.5mL of each sample. 2. To 0.5mL of your unknown samples (and dilutions of your samples), and to 0.5mL of the standards add 2.5mL of Lowry Reagent I. Mix well and let stand for 10min. The unknowns and the standards should be measured in the same experiment. 3. For each sample, add 0.25mL of Lowry reagent II and mix well immediately. Mixing right away is very important for successful results. Do not add the reagent to all of your tubes and then mix, because the local pH will degrade the Lowry reagent II if not 4. Measure the absorbance at 750nm (or with the Spec 20, at 650nm ) against a blank consisting of 0.5mL sample buffer (or water) processed as the samples
