Procedure Obtain a tablet of your unknown analgesic drug and samples (a few drops) of solutions of the three reference compounds. Record your unknown number. Crush the analgesic tablet on some filter paper, and place the powder in a 6-inch test tube. Add 1 mL of a 1:1 CH,CL,:methanol mixture, place the tube in a warm water bath in the hood, heat gently and stir the mixture for several minutes. Do not heat the solu- tion so as to cause the dichloromethane to boil away! While not all of the tablet will dissolve (the tablet may also contain insoluble binders and inorganic buffering agents), enough will go into solution to spot the TLC plate. Spot the known compounds and the unknown analgesic on a TLC plate as you did with the colored compounds. 1 = aspirin 2 = acetaminophen 3 = caffeine 4 = unknown Develop the plate using a solvent mixture having the composition: CH, Cl, :methanol:acetic acid (80:4:1). NOTE TO INSTRUCTOR: The 80:4:1 solvent ratio is critical. This solution must therefore be fresh to ensure the correct solvent composition. Otherwise, separa- tion will suffer. Examine the plate under UV light to see the components as dark spots against the fluorescent background. Outline the spots with a pencil. Then visualize the spots in an iodine chamber, as demonstrated by your instructor. Draw a replica of the plate in your Results section, Identify your unknown as you did with your colored unknown, Be sure to determine the R,'s of all compounds involved. If your unknown analgesic contains more than one compound, you should also run another TLC plate spotted with the unknown, each of the suspected knowns, and a mixture of the knowns prepared as before by spotting the knowns one over the other.
