In a chromatographic column, the concentration of a solute is expressed as the following. V4rDt „where c is the concentration, m is moles per unit cross-sectional area, D is the diffusion coefficient of solute, x is the distance along the column from the current center of the band (band center is always x-0), and t is time. The standard deviation of band is expressed as the following. (a) Prove that the peak width at half maximum is 2.350. (Hint: determine the maximum peak height using the first equation and then solve for x using the half of the maximum peak height). (b) Prove that the peak with measured at the baseline is 40. (Hint: determine the two tangential line equations at the inflection points of the first function above. Then, determine the crosspoints between the tangential lines and the baseline. )
